As far as I know, buses run only from Galati to Odessa, not from Bucharest; I could be wrong though and you could contact Bucuresti Filaret Bus Station for more accurate information:

Between Galati and Odessa cross Moldova and you need a Moldovan transit visa which you get in Bucharest. Without crossing Moldova, you could go Bucharest - Shmerinka by sleeper and then from there on by domestic UZ train:
Bucuresti Nord 07.40 - Shmerinka 08.57 R382/R60KJ (only sleepers)
Shmerinka 10.24 - Kuchurgan 15.43 R65MJ
Kuchurgan 16.57 - Odessa Golovna 18.15 R658ZJ
Or, more comfy, even though longer:
Bucuresti Nord 07.40 - Kiev Passajerskaya 13.46 R382/R60KJ (only sleepers)
Kiev Passajerskaya 17.28 - Odessa Golovna 05.27 R661KJ
